食管癌临床照射后细胞动力学改变
RADIATION-INDUCED ALTERATIONS OF DNA CELL CYCLE DISTRIBUTION IN ESOPHAGEAL CARCINOMA——Preliminary report
【摘要】 从1987年9月至1990年3月,共39例食管癌病人,随机分组后,20例接受术前放疗,19例单纯手术。术前放疗组接受5.2Gy/44h肿瘤量60Co单次照射。分析各种组织放射后细胞超微结构及周期进程变化,结果示。病人食管癌细胞的胞膜、胞核甚至细胞器均有一定程度的损伤表现,流式细胞检测(PCM)示食管癌原发肿瘤照射后G2细胞比例较照射前及未照射组均有明显升高,提示细胞周期进程受到了阻滞。同时比较原发癌与正常组织及癌转移淋巴结周期变化情况,发现放射对不同组织的细胞周期影响互不相同,至少不同步,并且个体病人间有明显差异。这可能试解释临床放射敏感性差异,并探索缩短常规术前照射的疗程。
【Abstract】 It remains abscure as to the radiation-induced changes of cell cycle status of either tumor or normal tissue in the human body. Due to the ethical reason, it is rather difficult to do such researches in patients. A randomized clinical trial in esophageal carcinoma treaed by surgery with or without irradiation was carried out on 39 patients, 20 of whom received combined surgery and radiation and 19, surgery alone. A single dose of 5.2 Gy (corrected) was given 44 hours before surgery by 300° rotation. The cell-cycle distribution was evaluated by flow cytometry. It was demonstrated that.there was a significant increase of cells in G2+ M phase after irradiation as compared to those before radiation or the control group. It could be speculated that it was due to G2blockage. A comparison of cell cycle status from primary tumor, normal tissue and lymph nodes showed that the radiation induced redistribution in these three tissues was different from each other. Further study will be carried out on the relationship between radiation-induced changes of cell cycle and clinical outcome of treatment.
