油菜抗病毒病与过氧化物酶关系
RELATIONSHIP BETWEEN VARIETAL RESISTANCE TO VIRUSES AND PEROXIDASE IN Brassica napus L.
【摘要】 用于叶期苗在不接种病毒条件下测试过氧化物酶同工酶结果,在相对迁移率17%—47%内,有6个位点出现酶带,抗、感病品种的酶带数量、酶带出现的位点和强弱程度有明显差别。高抗品种酶带数最多,有E3和E4强带,E3为弱带;高感品种酶带数最少,缺E6和E4,E3为强带;低抗和低感品种的酶带介于高抗与高感品种之间。用酶带数量和各酶带强弱程度,或用酶谱薄层扫描积分值计算距离系数,而后聚类,聚类结果与抗、感病性分组完全一致。证明抗病性与过氧化物酶同工酶有密切关系,用不接种苗酶谱测定可以代替接种鉴定和田间鉴定。
【Abstract】 The technique of polyacrylamide gel electrophoresis was employed to study the relationship between rapeseed varietal resistance to virus-PS and peroxidase in seedlings under the condition of noninoculation of viruses(TuMV.CMV).There were six loci of isozyme bands inrange of 17,-47% relative mobility(Rf)and obvious differences were observed between resistant and susceptible varieties in the number,presented locus and intensity of the bands.The number of the bands in highly resistant varieties with intensive bands.both E6(sixth locus)and E4(fourth locus),and with weaker E3 was more than that in highly susceptible varieties with intensive E3 and lacking E5 and E4.The isozyme zymogram of low resistant varieties or low susceptible varieties was simillar to either that of highly resistant varieties or highly susceptible varieties.Varieties were clustered by means of unweighted pair-group arithmetic average on basis of distance coefficient which was computed from the number and intensity of the bands,or from integral values of zymogram scan.The clustering results were completely consistent with grouping on basis of resistance and susceptibility.These results suggested close relationship between varietal resistance and isoperoxidase,and the measures of zymogram in noninoculative seedlings can replace inoculation or nature field trial to determine varietal resistance.
