长白蝮蛇毒纤溶组分的纯化及理化性质的研究
PURIFICATION AND CHARACTERIZATION OF THE FIBRINOLYTIC PRINCIPLE OF AGKISTRODON HALYS BREVICAUDUS
【摘要】 采用DFAE-Sepharose-CL-6B离子交换层析和Sephacryl S-200凝胶过滤层析,从长白蝮蛇毒中分离到五种具有纤溶活性的组分,并对其理化性质进行了较为深入的研究。结果表明,长白蝮蛇毒各纤溶组分具有纤溶酶和纤溶酶原激活因子的双重作用,能被金属酶抑制剂EDTA-Na2完全抑制。已纯化的三种纤溶组分,无出血活性。
【Abstract】 By means of DEAE-Sepharose CL-6B ion exchange chromatography and gel filtration chromatography, five different fibrinolytic principles were obta ned. Fraction Ⅱ, Ⅲ, Ⅳ appeared as single band by SDS-polyacrylamides gel electrophoresis.The molecular weight of fraction Ⅱ, Ⅲ, Ⅳ is 43 000, 17 800, 19 500 and isoelectic point is 4.66, 4.59, 4.44 separately. Five kinds of fibrinolytic principle show both fibrinolytic enzyme and plasminogen activator activity. Activity of five kinds of fibrinolytic principle was completely inhibited by EDTA-Na2. Hemorrhagic activity was not observed by animal test in Purified fraction.
