山莨菪碱对油酸所致离体肺损伤的防治作用及对氧自由基的影响
The effect of anisodamine on isolated lung injury induced by oleic acid and oxygen free radicals
【摘要】 用山莨菪碱(654-2)防治油酸引起的大鼠离体肺水肿性肺损伤,观察654-2对离体肺系数,肺通透指数,肺平均动脉压及肺静脉流出液中MDA,SOD,LDH含量的影响。结果表明:654-2可使离体肺系数及肺通透指数降低(P<0.01),MDA产生及LDH释放减少(P<0.01),SOD活性受到保护(P<0.01)说明654-2减轻肺损伤的作用与抗氧自由基有关。但进一步体外实验表明,各浓度654-2不能直接清除黄嘌呤—黄嘌呤氧化酶产生的氧自由基。上述结果提示,654-2可减轻油酸所致离体肺损伤,其作用可能在于稳定细胞膜。
【Abstract】 Anisodamine(654-2) was given to prevent oleic acid induced rat isolated lung injury. Changes in isolated lung coefficient, lung permeability index, pulmonary artery pressure and the content of malondiadehyde(MDA), superoxide dismutase(SOD), lactate dehydrogenase(LDH) in the effluence of pulmonary vein were observed. The results showed that while the lung was pretreated with 654-2, the lung coefficient, lung permeability index, content of MDA and LDH were decreased(P<0.01), and activity of SOD was protected(P<0.01). These results suggested that the protective effect of 654-2 may be correlated to scavenging of oxygen free radicals. But further experiments showed that 654-2 did not directly scavenge superoxide anion which come from xanthine/xanthine oxidase in vitro. It was therefore conclude that 654-2 possessed protective effect on oleic acid induced isolated lung injury, however the mechanism of 654-2 protection was not to scavenge free radicals directly but to protect the cell membrane.
