青椒根抑制血小板活化因子(PAF)活性成分的分离鉴定

【摘要】 自青椒(Zanthoxylum schinifolium Sieb.et Zucc.)根分离得到7个化合物,其中2个为新结构定名为青椒内酯(schinifolin I),乙酰氧青椒内酯(acetoxyschinifolin Ⅱ),其他5个为已知化合物,葡萄内酯(aurapten Ⅲ),白鲜碱(dictamnine Ⅳ),滨嵩内酯(scoparoneⅤ),茵芋碱(skimmianine Ⅵ),β-谷甾醇(β-sitosterol Ⅶ)。根据理化性质及光谱数据,Ⅰ,Ⅱ化合物鉴定为7氧代(3′,7′-二甲基-2′,6′-辛二烯)-8-甲氧基香豆素及7氧代(3′,7′-二甲基-5′-乙酰氧基-2′,6′-辛二烯基)8-甲氧基香豆素,化合物Ⅰ、Ⅱ、Ⅲ,Ⅴ在由血小板活化因子(PAF)引起的血小板聚集实验中,具有拮抗血小板聚集的活性。

【Abstract】 Two new compounds,schinifolin Ⅰ and acetoxyschinifolin Ⅱ,along with five known compounds(auraptenⅢ,dictamnine Ⅳ,scoparone Ⅴ,skimmianine Ⅵ,and β-sitosterol Ⅶ)were isolated from the roots of Zanthoxylum schinifolium Sieb.et Zucc.col- lected in Yixing County,Jiangsu Province.The structure determination was based upon spectroscopic analysis(UV,IR,MS,PMR,CMR,2D NMR).The structures of schinifolin Ⅰ and acetoxyschinifolin Ⅱ were elucidated as 7[(3′,7′-dimethyl-2′,6′-octadienyl)] oxy-8-methoxy-2H-1-benzopyran-2-one,and 7[(3′,7′-dimethyl-5′-acetoxy-2′, 6′-octadienyl)]oxy-8-methoxy-2H-1-benzopyran-2-one,respectively. In the test of platelet aggregation caused by PAF,compounds Ⅰ,Ⅱ,Ⅲ and Ⅴ showed inhibitory activity.
