异搏定影响交感神经节的突触后机制
POSTSYNAPTIC MECHANISMS OF VERAPAMIL ON EFFECTS IN SYMATHETIC GANAGLIA
【摘要】 本文采用细胞内记录技术,以压力注射仪喷注外源性乙酰胆碱(ACh)诱发的 ACh 电位为指标,观察了异搏定对离体蟾蜍椎旁交感神经节阻滞效应的突触后机制.结果表明,较高浓度(100μmol/L)异搏定引起膜轻度去极化并抑制 N 和 M 型胆碱能电位.这种作用似乎与异搏定的Ca2+通道阻滞效应无关,更可能是因突触后膜非失活性钾外向电流减小和非特异性地阻滞 Na+、K+通道所致.
【Abstract】 By using intracellular recording techniques,the experimental observations on the block effects of verapamil were made to investigate the postsynaptic mechanism of inhibiting synapic transmission.The experiment indicated that verapamil at higher concentration(100 μmol/L)can evoke a slight depolarization in the majority of cells(72%)and obviously depress acetylcholine(Ach)potentials induced by pressure injection of exogenous ACh.The effects of verapamil were concentration-dependent and not related to extracellular Ca~2concentration.It seems possible that verapamil has effects on reducing non-inactivation K~+outward-current (Im)and non-specifically depressing Na~+,K~+channels in postsynaptic membrane,and influ- ences synaptic transmission.
