大鼠腹膜细胞体内、外短期致癌试验——检测外用避孕药烷苯醇醚潜在致癌性

【摘要】 本文应用Nashed等人建立的大鼠腹膜细胞短期致癌试验法检测了现市场上应用的外用杀精子避孕药烷苯醇醚的潜在致癌性。实验结果:生理盐水、壬苯醇醚、以及烷苯醇醚的两个剂量(4 mg/kg和40 mg/kg)处理的大鼠,其腹膜细胞经体内外短期培养均未见有大于9个细胞的集落产生。而甲基亚硝基胍处理的大鼠,则可见9~30个细胞的集落(13.5±8.6个/20个高倍视野),且还可见100个细胞以上的巨大集落。依据判定指标表明烷苯醇醚无明显潜在的致癌作用。结合致畸和致突变实验结果证明该药是比较安全的。

【Abstract】 The carcinogenic potency of Alkyloxynol-741 (AP-741) was tested by using rat peritoneal cell short-term carcinogenic test method which was set up by Nashed.N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) was used as positive control. Saline and nonoxynon-9 (NP-9) were used as negative control. Two dose groups of AP-741 (4 mg/kg and 40 mg/kg)which were 4 times and 40 times the human’s doses were tested.The results showed that no colonies of more than 9 cells were seen in the saline and NP-9 groups and the two dose groups of AP-741. However, 12+8.3 colonies(9-30 cells/ colony) and 4.5 + 4.2 colonies (30~300 cells/colony) were seen in the MNNG group. According to the Nashed criterion, we can say MNNG has potential carcinogenesis, while AP-741 hasn’t any potential carcinogenesis and its use as a vaginal contraceptive drug is safe.
