甜瓜原生质体培养及胚状体发生的初步探讨
A PRELIMINARY STUDY ON THE PROTOPLAST ISOLATION CULTURE AND EMBRYOID GENESIS FROM Cucumis melo L.
【摘要】 以甜瓜无菌苗子叶为供试材料游离原生质体,原生质体在1/2MS培养基附加2,4-D1.0mg/L、6—BA0.5mg/L中做液体浅层培养。4~5天细胞第1次分裂,两周后形成小细胞团,添加低渗培养基继续培养,产生大量愈伤组织和部分胚状体。
【Abstract】 Protoplasts were isolated from cotyledons of Cucumis melo and cultured in 1/2MS (half strength macronutrients except CaCl2·2H2O) liquid medium supplmented with 2,4—D 1.0mg/L, 6—BA0.5mg/L. The first division was observed 4 days of culture. A great deal of calli were obtained and some small particles which were globular or heart-stage embryiods were formed. Then transfered to MS solid medium no hormone rhizogenesis occured.
