果蝇中E(w~a)及其对copia插入突变基因w~a的作用
ENHANCER-OF-WHITE-APRICOT AND ITS EFFECTS ON THE COPIA INSERTION ALLELE WHITEAPRICOT IN DROSOPHILA MELAGOGASTER
【摘要】 果蝇中,基因Wa是反转录病毒样转座因子copia对红眼基因W+的插入造成的。Wa果蝇的眼色为杏黄色。Wa果绳的大部分RNA产物都是终止在copia 的3’LTR,仅仅产生少量的结构正常的RNA。W?果蝇的眼色又受到了基因E(W?)的影响而变得更浅。Northern 分子杂交的结果表明,这种对色素的影响与W?果蝇的结构正常的RNA 的量减少是相一致的。用γ射线诱变得到3个E(W?)的回复基因,它们都是隐性致死,致死作用发生在幼虫阶段。我们认为,E(W?)是一个反效等位基因,其基因产物与野生型等位基因相互作用。野生型E(W?)+的基因产物对W?的前体RNA 的拼接起正向作用,或者对该RNA 的多聚A 加尾起负向作用。
【Abstract】 In Drosophila melanogaster,the w(?) al-lele differs from the red-eye gene(w~+)bythe insertion of the retrovirus-like transpo-sable element copia within the transcriptionunit.The w(?) flies have the apricot eye co-lor.Most RNAs derived from w(?) have 3’termini within the 3’ LTR of copia,andonly small amounts of structurally normalRNA are produced.The eye color of flieswith w(?) is affected by the gene Enhancer-of-white-aprocot [E(w(?))] and become ligh-ter.This effect on pigmentation is correla-ted with a corresponding decrease in whiteRNA having wild-type structure which wasdetermined by the Northern blot.Three re-vertant alleles of E(w(?)) was generated bythe irradiation with γ-ray.These allelesare recessive lethals with death occurringduring the larval stage.The original E(w(?))allele is an antimorph and produces a pro-duct that interferes with the activity of thewild-type gene which have the product ex-pected to either have a positive role in thesplicing of w(?) RNAs or a negative role inpolyadenylation.
