反相高效液相色谱法快速测定人尿中假尿嘧啶核苷

【摘要】 <正> 人尿中的修饰苷主要是由转移核糖核酸(tRNA)酶解产生的。现已发现:在人体癌变组织中从tRNA到假尿苷的转换率比周围正常组织高。癌症患者尿液中的修饰核苷比正常人的含量高,尿液中的修饰核苷以假尿苷含量最高,它因不被酶解而成为tRNA代谢的最终产物。基于以上特点,国外已有许多科学家
【Abstract】 A method for the rapid detemination of urinary pseudouridine by high performance liquid chromatography is described. In this method, the urine collected was stored at-20 ℃ for 24h and then thawed and centrifuged. The supernatant was loaded onto the column and the amount of pseudouridine was determined by measuring the peak area at 254nm. The detection limit of concentration was 3nmol/ml and the recovery was 100%. Furthermore, the determination of urinary pseudouridine could be used as a tumor marker in the follow-up of neoplastic disease and in monitoring therapeutic efficacy against cancer.
