细胞生物反应器大规模培养草鱼细胞和病毒

【摘要】 应用细胞生物反应器微载体悬浮培养法,对草鱼胚胎细胞 CP-80 进行大规模培养,同时增殖草鱼出血病病毒。细胞经培养6天后,细胞量可达到8.4×106cells/ml,较始接量(2×105cells/ml)提高了41倍。接入草鱼出血病病毒后,经5天培养增殖,毒力达到8.0LgTCID50/0.1ml,较始接毒力(3.75LgTCID50/0.1ml)增加了4.25个对数值。

【Abstract】 The embryo cell strain (CP-80) of grass carp (Ctenopharyngodonidellus) has been cultured on a large scale by using microcarrier suspension cul-ture. After being cultured with microcarriers for 6 days, the cell density can reachso about 8.4×105 cells/ml, which is 41 times as high as the initial inoculative con-centration (2×105 cells/ml). Afterwards, the haemorrhagic virus of grass carp isinoculated into the bioreactor while the cells have just been full on microcarriers.The titer of virus can be increased to 8.0 LgTCID50/0.1ml in 5 days incubation,which is 4.25 logarithm values as high as the initial virulcnce.
