茶叶抗氧化剂抗氧成分的分离和鉴定
ISOLATION AND IDENTIFICATION OF THE ANTIOXIDATIVE COMPONENTS IN TEA ANTIOXIDANT
【摘要】 茶叶抗氧化剂的初步分离采用葡聚糖凝胶 LH-20柱层析(洗脱剂无水乙醇,280nm 检测)得到12个部分,其中5个部分对亚油酸具有很强的抗氧化活性.采用高效液相色谱分析法测定了这5个抗氧性部分的组成.经过进一步层析分离,纯化后利用熔点测定、纸层析、气相色谱、高效液相色谱、红外光谱,质谱以及气-质联用分析等现代分析方法鉴定出1—(—)—表儿茶素、(—)—表棓儿茶素、(—)—表儿茶素没食子酸酯、(—)—表棓儿茶素没食子酸酯、d—(+)—儿茶素和咖啡因等6种抗氧成分。
【Abstract】 The tea antioxidant product was initially chromatographed on a Sephadex LH-20 column withethanol elution and detection at 280nm to obtain twelve fractions.Among them,five fractions showedstrong antioxidant activity toward the oxidation of linoleic acid.The composition of the five antioxidativefractions was determined by HPLC analysis.Six antioxidative components,1-(-)-epicatechin,(-)-epigallocatechin,(-)-epicatechin gallate,(-)-epigallocatechin gallate,d-(+)—catechin and caffeine,were isolated from the fractions by further chromatography,and were identified by MP,paperchromatography,GC,HPLC,IR,MS,and GC-MS methods after purification.
