苏云金杆菌187菌株的质粒检测和抗生素抗性研究

【摘要】 采用改良的酶碱综合法和琼脂糖平板电泳,测定187菌株具有11条质粒带;热处理表明至少含有8个质粒。抗生素抗性测定分析表明,187菌株为Amp(?), Str、Km、Gm、Cm、属抗药型,能分泌高活性的B-内酰胺酶。用0.04μg/mLEB42℃处理,筛选到4个稳定的187Amp(?)突变株。

【Abstract】 Plasmid detection and analysis of Bacillus thuringiensis subsp, israelensis 79-w-187 using the modified Birmborim and Doly procedure as well as Van Den Hondel’s method revealed the display of 11 plasmid electrophoretic bands on agarose gels and the presence of at least 8 plasmids. The test of antibiotic resistence showed the 187 strain was resistant to high levels(up to 2mg/ml)of ampicillin and sensitive to streptomycin, kanamycin, Sentamicin and chioromycetin. Four colordes of the 187 strain that were sensetive to ampicillin were isolated by incubating the bacterium at 42℃ for 24 h,the curing reagent was used at concentration of 0.04ug/ml ethidium bromide.
