烟曲霉变应原部分纯化和分析
Partial Purification and Analysis of Aspergillus Fumigatus Allergen
【摘要】 烟曲霉变应原经90%饱和硫酸铵盐析,将盐析液蛋白作Sephad-exG-200层析,出现三个洗脱峰。以相同蛋白浓度(6μg/ml)的粗制液、盐析液、上清液,及各段层析液对20例病人进行皮肤试验检测变应原活性,结果提示四种制备物均有活性,但层析液第二峰活性较高。用SDS-PAGE检测盐析液和各段层析液蛋白质分子量,结果表明主要含有38000、64000,其次为28000、44000、54000以及大于94000的蛋白质。
【Abstract】 Aspergillus fumigatus (A.F.) extract was salt precipitated by 90% saturated ammonium sulfate. The precipitated protein was isolated by gel filtration with sephadex G-200. Three elution peaks were observed. Twenty patients underwent Skin tests with crude extract, precipitated protein solution, supernatant and various fractions of ghromotography with the same protein concentration of 6μg/ml. Results showed that allergenic activities existed in ali of the preparations The activity of the second peak fraction was more potent. In SDS-PAGE, the molecular weight of proteins of various fractions were mainly 38000, 64000, and the nest were 28000, 44000, 54000 and above 94000. It was suggested that allergens of A.F. were widely distributed in various fractions with different MW outlined above.
