【Abstract】 Pollens of Ginkgo biloba L.(G. b.l.p) have been found to be a kind ofimportant allergen which causes pollinosisin Chengdu.The goal of thisstudy is to purifyG. b.l.p and to determine the allergenicityand immunogenicity of various fractions.Crude extract was purified by gel filtrationwith Sephadex G25, then G75.Two elutionpeaks were beserved. On SDS--PAGE, themolecular weights of protein of the 1st peakand the valley were 30--42 kd and 13--18kd,respectively, and that of the 2nd peat wasless than 13 kd. 40 patients with allergic rhinitis and/orasthma underwent the skin test with crudeextract and various fractions of gel filtra-tion it revealed that the strongest allergenicactivity existed in the 1st peak and therewas mild allergenic activity in the 2ndpeak. The in vitro allergenic activity andimmunogenic activity of various fractionswere examined by ELISA inhibition test.It was further confirmed that the allergenicactivity and immunogenic activity of the1st peak were the strongest, and those ofthe 2nd peak were the lowest. It is suggested that diagnosing reagentscan be made satisfactorily by partial puri-fication,i.e. discarding the inactive frac-tions, since allergenicity exists in variousfragments. But fractions of allergen withhigh IgG immunogenicity should be selectedto produce immunotherapy agents so as toenhance the production of blocking antibodyand thus improve the therapeutic effect.更多还原
