黄曲霉毒素B1人工抗原及抗体的制备研究

【摘要】 作者用黄曲霉毒素B1(AFB1)与羧甲基羟胺半盐酸盐(CMO)在碱性介质中回流反应,使AFB1生成AFB1羧甲基肟(AFB1-oxime),将此衍生物偶联到牛血清白蛋白(BSA)上,制得人工抗原AFB1-oxime-BSA。该人工抗原经纯化后免疫家兔,获得高效价(1:100 000)、高特异性的抗AFB1的抗体,可供用于免疫分析法测定AFB1的含量。

【Abstract】 Carboxymathyloxime of afla-toxin B1 (AFB1)was synthesized by refluxingAF B1 with carboxymethoxylamine hemi-hydrochloride for 4 hours in the presenceof pyridine. AF B1-BSA(antigen) was madesuccessfully by coupling AF B1-oxime withbovine serum albumin(BSA) in the presenceof a water soluble carbodiimide. The rab-bits were immunized with the artificialimmunogen which had been purified by di-alysis. The titers of antisera were dater-mined by indirect enzyme-linked immuno-sorbant assay (ELISA) which was develo-ped by ourselves. The highest titer was1: 100 000.
