肿瘤坏死因子和(或)平阳霉素对人食管癌细胞株(Eca-109)体内杀伤作用

【摘要】 作者用重组人肿瘤坏死因子α(rh-TNFα)和(或)平阳霉素(PYM)对人食管癌细胞株(Eca-109)裸鼠移植瘤的抗瘤作用进行了动态观察。结果证明,rh-TNFα局部和腹腔用药组之肿瘤体积比对照组明显缩小(P<0.05);组织学显示,瘤细胞变性,核固缩、核碎裂即凋谢(Apoptosis)明显增加,核分裂相减少;且有大片状出血坏死,癌巢周围大量中性粒细胞、淋巴细胞等浸润。提示TNFα对Eca-109移植瘤细胞具有明显抑制和杀伤活性,与PYM合用可显著增强其作用。其中,TNFα的免疫调节功能在机体抗肿瘤过程中可能亦起着重要作用。

【Abstract】 The antitumor effects of recombinant human tumor necrosis factor-α(rh-TNFα)and/or pingyangmycin were examined on human esophageal cancer cell line Eca-109 implanted subcutaneously in nude mice. The growth of the tumor was inhibited by TNFα administrated regionally or peritoneally for one to two weeks. The size of tumor in treated mice become significantly smaller than that in control ones (P<0.05). Histological examination showed that the tumor cells increased in numbers of pyknosis, karyorrhexis as well as apoptosis, decreased in mitotic activity and cytoplasmic degenerations. Partial tumor became hemorrhagic necrosis. Heavy infiltration of neutrophils, lymphocytes, etc, surrounding the nests of tumor ceils were found. It is suggested that rh-TNFα may have cytotoxic and cytostatic effects against the implanted Eca-109 cells in vivo. rh-TNFα may be a useful adjuvant to the drugs which have well-known anti-tumor activities. Besides, TNFα as an immunologic modulator may play an important role in the TNF-induced anti-tumor activities.
