敌枯双慢性毒性实验的病理学研究——大鼠甲状腺和肝超微结构的改变
Pathological Study of Chronic Toxicity of N,N~1-Methylene-bis-(2-amino-1,3,4,-thiadiazole)——Ultrastructural Changes of the Thyroid and Liver of Rats
【摘要】 本实验用敌枯双按不同剂量(1.0、0.2、0.05、0.01 mg/kg/d)配成不同浓度的溶液分别供4个剂量组的 SD 大鼠(每组30只)饮用,并设对照组(30只,仅饮用去离子水)。在第55周处死动物,每组随机选3~5只取甲状腺、肝脏作电镜观察。结果:4个剂量组大鼠甲状腺滤泡腔扩大,滤泡细胞变扁平,粗面内织网扩张呈囊状,线粒体肿胀,嵴变短而少,核膜内陷,异染色质凝集呈板或染色质边集;部分滤泡上皮细胞脱屑到滤泡腔内,可见畸形核、裸核及核碎裂。肝细胞线粒体呈“O”形,“C”形,粗面内织网呈囊状扩张,大滴脂肪变,核染色质边集,核均质化。实验表明敌枯双对甲状腺、肝组织均有毒性作用,超微结构改变程度与农药剂量呈正相关。
【Abstract】 In this experiment,rats were given 1.0,0.2,0.05,0.01,and 0 mg/kg.d of N,N~1-methy- lene-bis-(2-amino-1,3,4,-thiadiazole)by drinking respectively.All the rats were sacrificed in the 55th week.Thyroids and livers of 3 or 5 rats in each dose group were removed for electronic micro- scopic observation.The result showed that follicular cavities of thyroid were dilated,follicular cells became flat,rough endoplasmic reticulum revealed vacuolar,mitochondrias were swollen with short- ening cristae,nuclear membrane caved in,and heterochromatin presented a shape of board or chro- matin margination.Additionally part of follicular cells dropped to follicular cavities,nucleus of which were deformed,rude and broken.Mitochondria of liver cells presented a shapes of“O”or“C”,their rough endoplasmic reticulum was dilated,big drops of fatty were found in their cytoplasrna chromatin margination and nuclear homogenization were present.Changes of the ultrastructures proportionately increased with dosage of drugs.The result suggested that the drug be toxic to thyroid and liver of rats.
