性别决定基因PCR在性别鉴定中的应用
Application of Sry (sex-determining region of the Y chromosome) PCR to Sex Identification
【摘要】 用PCR技术扩增Y染色体特异的性别决定基因(Sry)区域中347bp的片段来鉴定小鼠性别,结果表明该方法具有简便、可靠和准确等优点。简要地讨论了影响性别决定基因PCR的关键因素。
【Abstract】 The sex of mice was determined by the PCR (Polymerase Chain Reaction) amplification of a DNAfragment of 347bp of Y-specific Sry(sex-determining region of the Y) sequence. With the two pairs ofprimers, including one pair of primers of myogenin (as control), to the same reaction tube, forty-fiverounds of amplification gave rise to both a 347bp (Sty) and a 246bp(myogenin) fragment in malesamples, while 347bp fragment was absent from female samples. The results in three different strains ofmice indicated that this technique had advantages in simplicity, specificity, and accuracy. The majorfactors for successsful PCR were discussed.
