提高牛成熟卵母细胞和体外受精胚胎质量的研究
Studies on Improving the Viability of Bovine Matured Oocytes and In Vitro Fertilized Embryos
【摘要】 牛卵丘细胞—卵母细胞复合体分别放入含有10%胎犊血清+10μg/ml LH+1IU/mlFSH(试验组1)、10%发情牛血清(组2)和仅含1O%胎犊血清(对照组)的TCM 199培养液中进行22—24小时体外成熟培养后,再与体外获能的牛精子体外受精。三组受精卵分别与牛颗粒细胞单层共同培养7—8天,试验组1和组2的囊胚发育率分别为12.7%和13.5%,与对照组(6.9%)相比均具有显著差异(p<0.05)。体外受精囊胚经分割后移给受体母牛,于1991年4月首次产下世界上第一头体外受精半胚牛犊。说明牛体外受精半胚仍具有令人满意的活力。
【Abstract】 Effects of hormones (LH and FSH) or estrous cow serum (ECS) on bovine oocytes maturation were studied. Cumulus-oocyte complexes were matured in TCM 199 supplemented with fetal calf serum (FCS, 10% v/v), LH (10μg/ml) and FSH (1 IU/ml) in Experiment 1,10% ECS in Experiment 2, and 10% FCS in Experiment 3, respectively. After being matured in 5% CO2in air at 39℃ for 22—24hr, the complexes were fertilized in vitro with frozen-thawed bovine sperms, which had been capacitated by cadium inophore A23187 for 6 hr. The zygotes were co-cultured with bovine granulosa cell monolayer for 7—8 days. The results show that the blastocyst rate in the presence of added hormones (12.7%, Exp. 1) or ECS (13.5%, Exp. 2) is significantly higher than that in the presence of FCS (13.5%, Exp. 2), which is also significantly higher than that in the presence of FCS alone (6.9%, Exp. 3). The results demonstrate a beneficial effect of estrous cow serum, characterized by an elevated concentration of LH and FSH, on ovine oocyte maturation and zygote culture in vitro. Four blastocysts of bovine in vitro fertilization (BIVF) were bisected through a simple bisection method. The 8 demi-embryos were inserted into 8 porcine empty zones and cultured in vitro for 2—3hr, then they were transfered to 4 recipients, resulting 1 pregnancy. As a result, the calf was born in Apr. 1991. The study shows a satisfactory viability of BIVF embryos even after bisection.
