MH134-88体外细胞系的建立及其生物学特性的研究

【摘要】 MH134-88是将用四氯化碳诱发成功的C3H/HeN系小鼠的腹水型肝癌株(MH134)经极限稀释及克隆化方法在体外培养,连续传代而建成的一个纯度较高的小鼠肝癌细胞系。MH134呈悬浮培养,生长迅速,特性稳定,迄今已传200余代。本文对其生物学特性进行了较系统的观察与研究。支原体检测阴性,具有较大推广应用的价值。

【Abstract】 A murine hepatoma cell line designated as MH134-88 has been established by limiting dilution and cloning method from the transplantable ascites MH134 tumor strain of syngeneic C3H/HeN mouse. It has been passed for more than 200 generations. It is a rapid growing suspension cell line. It multiplied approximately 15.6 folds on day 5 with mitotic index reaching 1.4% on the 4th day. Morphologically, most of the cells of the earlier generations were round in shape rather uniform in size but later on as the number of passages increased it showed marked polymorphism. It was a hypertriploid cell line with karyotype of stem line as 73, XY, +M1+M2+M4+2(M3+M5+M6). It exhibited the essential similar morphological, immunological and transplantable features as those of its originated MH134 tumor strain. MH134-88 cell line was easily to be cultured, passed, frozen and resuscitated. PPLO were not detected. So MH134-88 may be of value for practical purposes.
