肿瘤坏死因子体内抗肿瘤活性及其对巨噬细胞激活作用的关系
STUDY ON THE RELATION OF ANTITUMOR ACTIVITY OF TUMOR NECROSIS FACTOR IN VIVO AND ITS ACTIVATION EFFECTS ON MACROPHAGES
【摘要】 在体外细胞毒活性实验中,小鼠乳腺癌Ca761-86细胞对rTNF不敏感,而在体内,rTNF却可明显抑制Ca761-86细胞的生长。根据(1)rTNF在体外可直接激活小鼠腹腔巨噬细胞杀伤Ca761-86细胞;(2)在体内由rTNF诱导和激活的腹腔和脾脏中的巨噬细胞均可杀伤体外培养的Ca761-86细胞;提示rTNF在体内对Ca761-86细胞的抑制瘤效应与其对巨噬细胞的激活作用也可能有关。
【Abstract】 We investigated the cytotoxic activity of recombinant tumor necrosis factor (rTNF) against Ca761-86 mammary carcinoma both in vitro and in vivo. In direct cytotoxic assay, Ca761-86 cells showed no sensitivity to rTNF. However, rTNF was found to be highly efficacious against subcutaneously implanted Ca761-86 mammary carcinoma.In order to explain this discrepancy, we examined the indirect effects of rTNF on Ca761-86 cells particularly through macrophages, we found that rTNF can directly activated macrophage for killing Ca761-86 cells in vitro. In vivo, peritoneal and splenic macrophages induced and activated by rTNF also showed cytotoxic activity against Ca761-86 calls. This results suggest that rTNF, used in vivo, can induce and activate macrophages to kill tumor cells.
