灵芝抗衰老活性多糖GLA的化学研究

【摘要】 灵芝子实体经热水提取,乙醇分级沉淀,除游离蛋白质和小分子杂质,得到具有免疫调节作用及抗氧自由基活性的多糖GLA。进一步分离纯化,得到8个多糖均一体,对其中主要成分GLA4、GLA7、GLSP1~35个多糖均一体进行了化学结构研究。以完全酸水解、过碘酸盐氧化、Smith降解、光谱测定及气相色谱分析等证明,GLA4、GLA7为以葡萄糖为主的中性杂多糖,GLSP1为鼠李半乳聚糖肽,GLSP2及GLSP3为葡聚糖肽。它们的精甙键类型均以β-(1→3)(1→6)及(1→4)为主,GLA4尚有α-甙键存在。

【Abstract】 The polysaccharide GLA was obtaiaed from the fruit-body of Ganoderma lucidum through the ext-raction of hot water, fractional precipitation with ethanol, cleavage of the free protein and removal ofmicromolecular impurities. Immune regulatory function and SOD activity were fouad in GLA. Furtherisolation and purification yielded eight homogenous polysaccharides, five of them, CLA4, GLA7, GLSP1-3 accordingly, underwent structural studies. Through the procedure of total acidic hydrolysis, perio-date oxidation, Smith degradation, spectral determination and GC analysis, GLA4 and GLA7 were sho-wn to be heteropolysaccharides containig mainly glucose. GLSP1 to be a rhamnogalactan peptide. whileGLSP2 and GLSP3 to be glycan peptide.β-(1→3)(1→6)and(1→4)linkage are the main type of bond ex-isting in the molecules and α-bond is also existed in GLA4.
