表达狂犬病毒糖蛋白的重组痘苗病毒的组建与鉴定

【摘要】 把狂犬病毒标准攻击株(Standard challenge virus)的糖蛋白编码序列插入天坛株痘苗病毒的胸腺核苷激酶(TK)基因区,并置于痘苗病毒7.5k蛋白启动子(P7.5)的控制之下,构建了两株重组痘苗病毒。它们在起始密码子上游一侧的核苷酸序列稍有差别,但均能有效地表达狂犬病毒的成熟糖蛋白并能转运至细胞膜上。免疫学试验及动物试验结果表明,重组痘苗病毒表达的狂犬病毒非融合性糖蛋白分子量约为64kD;均能与狂犬病毒天然糖蛋白单克隆抗体发生特异性反应,并能在小鼠和家兔中诱生高滴度狂犬病毒中和抗体(VNA)并对致死量狂犬病毒攻击动物具有保护能力。

【Abstract】 Two recombinants of vaccinia virus ( Tian Tan strain ) containing the coding sequence of Challenge Virus Standard ( CVS ) rabiesvirus glycopro tein were produced by insertion of the glycoprotein gene into the vaccinia tk gene. The coding sequence was controled under the vaccinia early -late promoter p7.5. The recombinants differed slightly in its sequence upstream of the translation start codon and both express effectively mature glycoprotein of rabiesvirus, which was transported to the cell membrane.Immunological assays and trial immunizations in animals showed that the molecular weight of non-fusion glycoprotein of rabiesvirus approximates 64kD. It react specifically with antibody against authentic glycoprotein of rabiesvirus. Each recombinant induced rabiesvirus neutralizing antibodies in mice and rabbits and protection against lethal street virus challenge.
