草鱼出血病病毒的糖蛋白和结构多肽的抗原性

【摘要】 研究了草鱼出血病病毒(GCHV)武汉南湖株的糖蛋白,初步确定5种结构多肽含糖成分,它们分别是Vp130、Vp120、Vp113、Vp107和Vp42。病毒多肽经SDS-PAGE后,可完全从凝胶转移到硝基纤维素膜上。同时也证实了该病毒有11种结构多肽。蛋白印迹-ELISA法检测兔抗GCHV多肽的抗体仅有7种成分,它们对应的多肽是Vp130、Vp120、Vp113、Vp107、Vp65、Vp60和Vp52。将完整病毒裂解成多肽免疫家兔获得的抗血清,其主要有效成分为大分子量的主要多肽的抗体。

【Abstract】 It had been reported that the GCHV ( grass carp hemorrhage virus ) isolate from the South Lake in Wuhan has 11 polypeptides and six of them are the components of the viral core by our laboratory. We did further experiments on the viral structural polypeptides. By PAS reac-, tion, it was shown that polypeptides Vpl30, Vpl20, Vpll3, Vpl07 and Vp42 are glycoproteins. Eleven structural polypeptides, namely Vpl30, Vpl20, Vpll3, Vpl07, Vp75, Vp65, Vp60, Vp52, Vp42, Vp39 and Vp31 were demonstrated by amino-black staining of transferred strip. A rabbit antiserum against GCHV polypeptides were prepared by our laboratory. The Western-blot-ELISA technique was applied to analyse the antigenicity of the viral structural polypeptides. Antibodies against Vpl30, Vpl20, Vpll3,Vpl07,Vp65,Vp60 and Vp52 were demonstrated in the rabbit antiserum. The major effective components of the rabbits antiserum are antibodies against the polypeptides with large molecular weights of the virus.
