实验性家兔颈动脉冷冻保存与移植
EXPERIMENTAL CRYOPRESERVATION AND ALLOTRANSPLANTATION OF CAROTID ARTERIES IN RABBITS
【摘要】 本实验以15%二甲基亚砜(DMSO)作为低温保护剂,采用两步冷冻步骤,选择4种不同的冷冻维持温度(-20℃、-40℃、-60℃、-80℃)对家兔的颈动脉进行冷冻,在液氮中保存48h后,施行同种异体移植。通过四唑盐还原试验、透射电镜、血管造影以及病理学检查,探讨冷冻的维持温度对血管的影响。结果表明,经-60℃维持温度冷冻后再保存于液氮中的异体血管,其代谢活性和超微结构得到部分保持,移植后排斥反应减弱,90天通畅率为87.5%,无动脉瘤发生。实验采取雌雄配对的方式,对血管内皮细胞的性染色质(Barr氏小体)进行检测分析,提示了内皮细胞的免疫排斥与修复保持相对动态平衡是维持异体血管移植后远期通畅的主要原因。
【Abstract】 With 15% dimethylsulfoxide(DMSO) as a cryoprotective agent,rabbit carotid arteries were stored in liquid nitrogen 48 hours by a two-step freezing schedule, in which different holding temperatures - 20℃, - 40℃ , - 60℃ and - 80℃ were employed in four groups and interposed in femoral artery as allografts. On the basis of tetrazolium test,transmission electron microscope,an- giography,and pathologic examination,the present study was undertaken to assess the effect ofholding temperature on cryopreserved arterial allografts. The results obtained suggest that at the holding temperature - 60℃ the metabolic activity and ultrastructure of allografts were partly preserved with a decreased immunologic reaction. There was a 87. 5 % patency rate at 3 months for such allografts with no evidence of aneurysm formation. With female arteries transplantation into male rabbits,sex chromatin bodies(Barr bodies) analysis suggests that the maintenance of long-term patency may be referable to a tendency of equilibrium between the rate of endothelial rejection and the rate of repair.
