凡拉帕米及其与米托蒽醌合用对裸鼠实验性Lewis肺癌转移的抑制作用
INHIBITION OF EXPERIMENTAL TUMOR METASTASIS BY THE CALCIUM ANTAGONIST VERAPAMIL AND ITS COMBINATION WITH DHAD
【摘要】 通过裸小鼠尾静脉内注入Lewis肺癌细胞造成实验性肿瘤肺转移,在体内研究了钙拮抗剂凡拉帕米[VP]及其与抗癌药米托蒽醌[DHAD]合用对转移性肺结节形成的影响。发现VP及其与DHAD合用均可明显减少Lewis肺癌细胞转移性肺结节的形成。体外血小板聚集试验亦证实钙拮抗剂VP可以抑制Lewis肺癌细胞诱致的血小板聚集。结果提示,VP有希望成为预防肿瘤转移的良好好辅助药物。
【Abstract】 We examined the Verapamil (VP) for its ability to inhibit met-astases in vivo and tumor cell-platelet interactions in vitro. In vivo, VP inhibited the experimental metastasis (pulmonary tumor colony formation, PTCF)induced by the intravenous injection of Lewis lung cancer cell suspensions into nude mice. The inhibitory rate of PTCF was 49.1%. When VP was given with DHAD,the inhibited rate of PTCF was increased to 66.7%. In vitro, VP inhibited Lewis cancer cells induced platelet aggregation in a dose-dependent manner. The results proved VP has antime-tastatic effect and might become suitable adjuvant for antimetastatic treatment in human cancers.
