甲氨蝶呤明胶微球肝动脉灌注后的药代动力学研究
PHARMACOKIN ETIC STUDY OF GELATIN MICROSPHERES CONTAINED METHOTREXATE AFTER HEPATIC ARTERIAL INFUSION
【摘要】 作者采用国内首创的新型化疗栓塞剂——甲氨蝶呤明胶微球(MTX—ms)和普通甲氨蝶呤(MTX),按MTX1.5mg/kg的量经大白鼠肝动脉灌注,然后从大白鼠肝静脉抽血测定血药浓度,以观察其药代动力学变化,作为临床应用MTX—ms治疗肝癌的前期研究。结果用MTX—ms灌注后MTX的生物半衰期和平均驻留时间较单纯MTX灌注明显延长;MTX的全身分布减少43%以上。由此可见MTX—ms具有药物缓释特性,并能将高浓度的MTX限制在肝脏内,减少和延缓MTX的全身分布。作者对其临床意义进行了讨论。
【Abstract】 Methotrexate Gelatin Microspheres (MTX-ms), an initial domestic newchemoembolization agent, and ordinary Methotrxate (MTX) were infused into hepatic artery of rats in quantity of MTX 1.5 mg/kg, then colect the blood sample from hepatic vein to determine the blood concentration of MTX, as a preclinical study of the treatment of hepatic carcinoma with MTX-ms. The result shown that after infusion of MTX-ms the biological half-time and mean resident time of MTX were much longer than that infusion of ordinary MTX alone, and wholebody distribution of MTX decreased by more than 43% . It is concluded that MTX-ms possesses properties of drug sustained-release and restricts the high concentration of MTX in the liver, reduces and slows down the wholebody distribution of MTX. Author has discussed it’s clinical significance.
