亚基特异性铁蛋白单克隆抗体对人肝癌与肝脏铁蛋白生化免疫性质分

【摘要】 <正> 1975年,Alpert首次报道肝癌组织铁蛋白等电聚焦电泳位置与正常肝有所不同,其等电点略偏酸性,而与人胎肝铁蛋白相似,称之癌胚铁蛋白(Carcino-fetal ferritin)。最近我们与意大利米兰大学生物医学技术部合作,以重组的H和L铁蛋白亚基及其相应的单克隆抗体与从人肝癌和肝脏中提取纯化的铁蛋白及其相应的多克隆抗体进行生化免疫性质比较,结果报道如下。

【Abstract】 The immunospecificity of ferritin from human hepatoma and liver has been studiedby H and L subunit specific monoclonal antibodies. SDS-polyacrylamide gel electro-phoresis showed that both hepatoma and liver ferritin were predominant with L-subunit. Ouchterlony test and western blot analysis demonstrated that both H and Lsubunit specific monoclonal antibodies can bind to ferritins from hepatoma and liver.Conversely, polyclonal antibodies developed aganist hepatome and liver ferritin respec-tively can only react with L subunit but not with H subunit ferritin.
