丹皮酚抑制培养主动脉平滑肌细胞的增殖及抗自由基作用
Paeonol inhibit the Proliferation of aortic smooth muscle cell in culture and the production of free radicals
【摘要】 用结晶紫染色法及3H—TdR参入法研究丹皮酚对培养兔主动脉SMC增殖的影响,结果表明,细胞生长第6天加入不同剂量的丹皮酚(10~160μg/ml)能显著抑制细胞增殖及DNA合成,量效关系明显。用黄嘌呤—黄嘌呤氧化酶—鲁米诺(luminol)发光体系探讨丹皮酚的抗O2自由基作用,结果显示丹皮酚能抑制细胞内2自由基的产生。
【Abstract】 This paper report the effect of paeonol on proliferation of cultured aortic smooth muscle cell (SMC) by means of staining cell nuclei with crystal violet and[3H]-TdR incorporation. When paeonol(10-160μg/ml media) was added at the d6of SMC growth, proliferation and DNA synthesis of SMC was significantly restrained, inhibition was increased as dose of paeonol enhanced (r=-0.99) . With chemiluminescence method, we also studied the role of paeonol in the production of free radicals. The results showed that paeonol might abstain free radicals formation by directive action or increase of superoxide dimutase activity in SMC. These evidence suggest that paeonol might inhibit the atherosclerotic plaque formation in cholesterol-fed rabbit by restrain the proliferation of SMC, and decrease the free radicals formation by increase the activity of superoxide dimutase.
