CdMT诱发大鼠肾损害的机理研究

【摘要】 经皮下给大鼠注射镉金属硫蛋白(CdMT,0,0.5,1.0mgCd/kg)诱发急性中毒作为慢性镉中毒的模型,观察中毒大鼠肾皮质的组织病理学改变和脂质过氧化作用(LPO),以及尿液主要生化指标的变化。结果发现,尿总蛋白、葡萄糖、γ-谷氨酰转移酶、碱性磷酸酶、N-乙酰-β-D-氨基葡萄糖苷酶含量均明显增加。光镜和电镜下见近曲小管细胞浊肿、水样变性、微绒毛稀疏、脱落,胞浆空泡和线粒体肿胀变性。肾皮质GSH和MDA的减少和增高与尿γGT和NAG有显著相关,提示LPO可能是CdMT的肾损害机理之一。

【Abstract】 With acute renal damage induced by a single subcutaneously administered cadmium-metallothionein(CdMT)in male Wistar rats was used as a modle for chronic Cd poisoning, the experiment was performed to search the effects of lipid peroxidation(LPO)on renal damage of CdMT. The morphologic changes in renals of intoxixicated rats were found as a widerspread vesiculation of proximal tubular cells with mitochondrial alterations and choronic retrograde degeneration,and urinary biochemical indicators for renal tubular injury and dysfunction were determined in the animals. The levels that renal glutathione (GSH) was significantly decreased and malondialdehyde (MDA) increased,which keep well correlationships with that of rGT and NAG in urine of rats injected with CdMT, suggesting that LPO is partly involved in the tubular damage induced by CdMT.
