柔红霉素的发酵研究
STUDY ON THE FERMENTATION OF DAUNORUBICIN
【摘要】 利用TLC、HPLC技术鉴定了柔红霉素产生菌SIPl 1482发酵过程中间产物ε—紫红霉酮、副产物13—双氢柔红霉素、13—双氢柔红霉酮,柔红霉酮、7—脱氧—13—双氢柔红霉酮和7—脱氧柔红霉酮等组份的变化。研究了外源添加试验,发酵条件包括发酵培养pH、温度及通气等条件对柔红霉素生物合成的影响,对副产物及中间体ε—紫红霉酮的转化。玉米粉—麸皮培养基能提高柔红霉素发酵效价,且降低发酵过程中副产物的生成。 在高氏一号合成培养基上不产柔红霉素的突变株SIPI 1482 NS-4通过加入玉米粉或玉米粉酸解液、或经纯化的酸解液组份1-B,能够使其恢复产生柔红霉素。在柔红霉素生产菌株SIPI1482和SIPIPF-25原来的玉米粉—麸皮培养基中添加一定浓度的玉米粉酸解液能够提高柔红霉素发酵效价10%和30%以上。
【Abstract】 The biosynthesis pathway of daunorubicin producer SIPI 1482 was proposed after carefully analysing the changes of various compounds and combining the tests for adding some precursors to the mutant or parent strain,,Through carefully analysing the results obtained from cultivating SIPI-1482 in different fermentation conditions, the parameters such as the constitution of medium, pH, temperature and aeration could affect not only the production of daunorubicin. but also the byproducts including the conversion of ε-rhodomycinone to its glycosides. The production of daunorubicin would increased and the byproducts would be decreased if the strain was cultivated in the medium containing 7% of corn meal.A daunorubicin-negative mutant SIPI1482-NS-4, which could not produce daunorubicin in Gause I medium, was recovered its daunorubicin’ s synthesis ability in the medium containing corn meal or its hydrolysate. when cultivating the daunorubicin producing strain SIPI1482 and SIPIPF-25 in the medium containing 7% of corn meal together with 1% of its hydrolysate the production of daunorubicin was increased more than 10 and 30 percentage respectively. The function of regulatory factor involved in daunorubicin biosynthesis was also discussed.
