氯霉素血浓度的高效液相色谱法监测

【摘要】 一种监测氯霉素血药浓度的高效液相色谱法,分析条件如下:分析柱:μbondpakC18;流动相:甲醇:1%乙酸(45:55);流速:1.0ml/min;检测波长:UV285nm,本方法样品峰分离好,保留时间为7.2min。峰高与浓度(1~50mg/L)呈线性(r=0.9999);血清提取回收率高,平均为92±3%;重复性好,变异系数<2.4%;正确性高,9个血清样品的测得浓度与配制浓度的差异为3.7±2.9%,特异性高,17种抗菌药物对氯霉素样品峰无干扰。本方法简单、灵敏、可靠,完全能满足临床对氯霉素血液及其他体液浓度的监测。

【Abstract】 A high-performance liquid chromatographic (HPLC) method formonitoring the blood concentration of chloramphenicol was described. The conditions of HPLC were as follows. Columns:μbondpak C18; mobile phase smethanol : 1 % acetic acid (45:55) ; flow rate:1.0 ml/min; detection: U.V. absoption at 285 nm. The peak of chloramphenicol was separated well with retention time 7.2 min. The stand, curve between peak hight and concentration was up to 50 mg/L (r = 0.9999) . Recovery from serum was 92 + /-3%. Reproducibility was good,coefficients of variation being<2.4%. Accuracy was determined by 9 prepared samples, the results obtained were 3.7 + /-2.9% of the assigned concentration. Specificity was good, none of 17 tested antimicrobial agents intefered. This method of assay thus appears simple, sensitive and reliable for clinical therapeutic monitoring chloramphenicol in serum and in other body fluids.
