恶性肿瘤患者染色体不稳定性的研究

【摘要】 对55名恶性肿瘤患者和27名正常人进行了淋巴细胞染色体畸变率(CAR),脆性位点(fra)表达串和SCE率分析,结果分別为8.89%,5.72%和9.64±2.35次/细胞(P<0.01),说明恶性肿瘤患者染色体稳定性较差,可导致肿瘤的发生。在CA所累及的441个位点中,284个与fra一致,317个与癌断裂点、127个与癌基因一致或邻近,说明fra癌断裂点和癌基因有一定关系。

【Abstract】 The rates of chromosome aberration, fragile site expression and SCE were analysed in lymphocyts of 27 normal controis and 55 patients with malignant neoplasms. Of the 141 sites that were involved in chromosome aberrations, 284 were fragile sites, 317 were at or near the loci of cancer break points and 127 at or near the loci of oncogenes. This indicated that the fragile sites, cancer breakpoints and cncogenes were associated with cancer development, perhaps some fragile sites were liable to chromosome breakage and rearrangement whick reactivated oncogenes and thus led to caucer development.The rates of chromosome aberration, fragile site expression and SCE of the patients were 8.89%, 5.72% and 9.64+ 2.35/cell, respectively, and those of the normal controls were 2.22%, 1.87% and 5.77+1.26/cell respectively. The differences in rates of chromosome aberration fragile site expression and SCE between the patients and the normal controls were very significant (P<0.01). This. suggested that the chromosomes of patients with malignancy were instable and predisposed to the develogment of cancers and this chromosome instability could be tested by cytogenetic technique.
