用放射自显影方法研究正常大鼠肾脏心钠素受体

【摘要】 用含123Ⅰ心钠素的Tris·HCl缓冲液孵育10μm厚的正常肾脏切片20℃1h,然后将切片暴露于X线片和核乳胶2~3周。结果显示,放射自显影标记银粒在肾皮质内呈弥漫性斑点状分布,而在髓质分布很少,显微镜下观察银粒在肾小球内呈高密度分布(37.2±7.5/100μm2),在肾小管则分布密度较低(17.1±3.8/100μm2)。资料提示:心钠素受体通过增加肾小球滤过率发挥利钠和利尿作用。

【Abstract】 The purpose of this paper was to study the autoradiographic localization of ANP receptors in normal rat kidney. Autoradiograms were generated by incubating 10μm sections of normal rat kidney with 125I~ANP in tris.HCL buffer for 1 hour at 20℃.Tissues were exposed to X film and emulsion for 2 to 3 weeks.The labelling in the kidney showed a diffuse granular pattern in the cortex with lower grain densities in the medulla.Sections exposed to the emulsion demonstrated that the labelling in the cortex was localized to grome-ruli,and a very high density of silver grain distribution was found in over-lying glomeruli (37.2 ±7.5/100μm2), and a lower density in the renal tubule (17.1±3.8/100μm2) .Our data suggest that the ANP receptors exist mainly in glomeruli and may increase the glomerular filtration rate, playing its role of potent natriuresis and diuresis.
