HSV体外感染对IgG产生的抑制作用和TNF及r-IFN对它的影响
Inhibitory Effect of HSV Infection in Vitro on IgG and Effect of TNF and r-IFN on It
【摘要】 本文观察了体外HSV-I感染对人淋巴细胞增殖、分化,产生免疫球蛋白的影响。HSV-I能感染PWM刺激的人扁桃体淋巴细胞,并抑制其增殖和免疫球蛋白(IgG)的产生,感染实验组的IgG产量较对照组明显降低(P<0.005)。HSV-I感染后,上清中可检出一定量的病毒,其滴度为103·2—105·7TCID50/ml;直接免疫荧光法检出病毒抗原阳性细胞为3%—8.5%。在感染实验系统中加入重组的IFNa(r-IFNa)、IFNγ(r-IFNγ)和纯化的肿瘤坏死因子(TNF),观察到r-IFN。和TNF均有一定程度抗病毒作用,部分解除HSV-I对IgG产生的抑制作用。和对照组相比,差异显著(P<0.01)。r-IFNγ对HSV-I的感染和IgG产生几乎无作用(P>0.05),但r-IFNγ对TNF有协同作用:抑制病毒增殖和解除HSV-I对IgG产生的抑制,比单独用TNF组更为显著(P<0.01)。
【Abstract】 We successfully established a system in vitro that HSV-Ⅰ infected human tonsillar lymphocytes. Tonsillar lymphocytes prestimulated with PWM(10μg/ml) were infected by living HSV-Ⅰ, so as to their proliferation and IgG synthesis were abated significantly(P<0.005).In the supernatants, the infective titers ofHSV-Ⅰ were about 103·2—105·7 TCID50/ml,number of viral antigen positive cells about 3%—8%.We found that some lymphokine and monokine can dampen the inhibition of infection of infectious HSV-Ⅰ The effect of purified recombinant IFN-α was stronger than that of IFN-γ(p<0.01). The purified tumor necrosis factor (TNF)also abated alone the inhibition of HSV-Ⅰ markedly, so that IgG synthesis enhanced more highly than that without adding TNF (P<0.01). TNF in combination with IFN-γ caused a larger increasing in IgG synthesis than that observed for TNF alone(P<0.01)or IFN-γ alone(P<0.01). This result showed that the activity of TNF had dramatically synergized with IFN-γ.
