异尖线虫四种抗原的酶免测定法检测抗体的比较
Comparison of Four Different Kinds of Anisakis Larvae Antigen for Circulating Antibody Detection by ELISA
【摘要】 以异尖线虫幼虫(Anisakis type Ⅰ)实验感染家兔,用不同方法制备四种抗原,按不同感染强度和不同时期进行ELISA测定。四种抗原不同感染强度所得血清内均可测到特异性IgG抗体,但存在特异性抗体出现时间快慢和抗体水平高低问题。SEC抗原在其敏感性和制备等方面均优于其它三种抗原,感染1周时出现强阳性反应,且维持时间长,当阳性判断标准定为0.987(X±3SD)时,所有感染后血清对于SEC抗原和WWE抗原敏感性分别为75%和62.5%,感染早期(7d)开始OD值明显上升,维持时间短的为2周,长的为5周,当10周时恢复到感染前水平,说明具有早期诊断意义。
【Abstract】 Specific. IgG antibody was demonstrated by the ELISA in the sera of rabbit infected with Anisakis type I according to four Various antigens, different intensity and course of infection. SEC antigen was superior to other three antigens in sensitivity and preparation The OD values (opitical density) reached plateau at 1 week and maintained long time. It was the most significant in diagnosis. When 0.987(Mean±3SD) were considered as cut-off value, secretory-excretory (SEC) antigen and whole worm extract antigen in all of the sera after infection were 75% and 62.5% respectively, but there was false negative reaction. The OD values of SEC antigen increased gradually at early period (one week) and maintained from 2 to 5 weeks, then restored to level before infection at 10 weeks.
