应用微量细胞培养间接免疫酶组化法检测单纯疱疹病毒的实验研究
AN EXPERIMENTAL STUDY ON THE USE OF MICRO-CELL CULTURE TECHNIQUE ( MCCT ) AND IMMUNOPEROXIDASE ASSAY ( IPA ) FOR DETECTING HERPES SIMPLEX VIRUS ( HSV )
【摘要】 报道了微量细胞培养间接免疫酶组化法检测感染细胞内单纯疱疹病毒的实验研究。用燃烛干燥缸代替CO2培养箱培养细胞,以多种敏感细胞培养病毒及不同固定液固定细胞等作了比较观察,并初步探讨了影响试验的因素,确立了最佳实验条件。实验结果表明此法敏感性高、特异性强、操作较简便,可望为临床诊断实验室提供一种快速检测病毒的方法。
【Abstract】 In this study, the cells sensitive to HSV infection were cultured in the 40-hole polyethylene plate, which was placed in the sealed candle jar and incubated at 37℃ to substitute for the use of conventional CO2 incubator. Cells of various origins and different fixative liquids were compared and the other influential factors were studied to set the optimal experimental conditi-ons. The established procedures were used to detect HSV and the results show that this simplified method is highly sensitive and specific, and therefore may serve the ordinary chinical laboratory as a rapid means for detecting viral pathogens.
