人胃粘膜细胞器标志酶电镜细胞化学研究

【摘要】 应用电镜酶细胞化学方法对15例人体正常胃粘膜细胞碱性磷酸酶(AlP)、酸性磷酸酶(AcP)、葡萄糖-6-磷酸酶(G6Pase)、焦磷酸硫胺素酶(TPPase)及细胞色素氧化酶(CCO)进行了超微结构水平的研究,并与光镜结果进行了对比观察。发现正常胃粘膜上皮细胞无AlP反应;AcP仅见于主细胞的胞溶酶体及分泌溶酶体内;TPPase定位于胃粘膜表面上皮细胞及主细胞高尔基体反面第1~3层膜囊、GERL及少数内质网中;G6Pase和CCO见于所有胃粘膜上皮细胞内,但G6Pase反应以主细胞显著,CCO反应以壁细胞显著,与其细胞功能相一致。作者认为电镜酶细胞化学研究能更真实地反映出细胞的功能活动和代谢状态。

【Abstract】 Ultrastructural localization of A1P, AcP,G6Pase,TPPase and CCO in 15 cases of normal human gastric mucosa were investigated with electron microscopic cytochemical technology. The results showed in normal gastric epithelium there is no A1P reaction.The AcP reaction product is only localized in autolysosomes of chief cells with the lysosomes showing active crinophagy.The TPPase reaction product is localized in the cisternae 1-3 stacks of the trans side of Golgi apparatus, GERL and a few endoplasmic reticulum. The G6Pase and CCO reaction products are found in both endoplasmic reticulum and mitochondria in normal gastric epithelium, but the former is more apparent in chief cells and the latter in parietal cells, which is consistent with their functions respct-ively. The biologic significance of the organelle marker enzymes were also disccused. The authors consider that ultracytochemistry can more really reflect the functional activity and metabolic characters of gastric epithelium.
