大鼠鼻粘膜上皮细胞微核试验

【摘要】 本文报告了大鼠鼻上皮细胞微核试验。分离的鼻道粘膜以链霉蛋白酶和胶元酶消化得到上皮细胞,细胞产率为每只大鼠3.1×10~6,细胞存活率为71.6%。在涂片上,大鼠鼻上皮细胞可分为纤毛细胞、杯状细胞及其它细胞。大鼠鼻粘膜上皮自发微核率为1.76±1.2‰。以滴鼻法亚急性染毒,甲醛和二甲基亚硝胺可显著增加大鼠鼻上皮细胞微核率,并有剂量-反应关系。

【Abstract】 The micronucleus assay in rat nasal epithelial cells ( NEC ) was reported in this paper. The isolated turbinates were digested with pronase and collagenase to get single NEC. Mean cell yield per rat was 3.1×105 cells and cell viability was 71.6%. In smear, NEC were divided into three groups: ciliated cells, goblet cells and other cells. Sponteneous micronucleus frequency in rat NEC was 1.76±1.20‰. Using the subacute treatment of nasal drip formaldehyde and dimethylnitrosamine induced the increases in NEC micronuclei in a dose related manner.
