6巯基鸟嘌呤和干扰素对HL-60细胞中相关基因mRNA表达的调控
Effect of 6-thioguanine and interferon on related gene mrna in hl 60 cells
【摘要】 为研究6-巯基鸟嘌呤(6—TG)和干扰素(IFN—α和IFN—γ)对HL—60细胞中相关基因表达的调控作用及其与细胞增殖和分化的关系,实验观察了这两类因子对HL—60细胞的生长抑制作用,分化诱导作用及其对C—myc原癌基因mRNA和干扰素相关基因pIFN—IND-2的56KmRNA表达的调控作用。 结果表明:对于HL—60细胞的克隆生成,6—TG是强烈的抑制剂,而干扰素类几无抑制作用。在NBT实验中,这两类因子都产生了类似程度的诱导HL-60细胞向粒细胞方向的终末期分化。对Northern Blot的结果分析发现它们对相关基因产生了相似的调控作用。C—myc mRNA早期的增加和后期的明显减少;56K mRNA则有程度不同的增加现象。 结果提示:在HL—60细胞中C—myc原癌基因与其分化密切相关,pIFN—IND—2基因也可能与之有关。对相关基因的类似调控作用可能是6—TG与干扰素联合应用的分子生物学基础。
【Abstract】 In order to study the relationship between some related genes’ expression and cell proliferation, cell differentiation, the effect of 6-thioguar-ine (6-TG), interferons (IFN-α and IFN-γ ) on HL-60 human leukemia cell growth, cell differentiation and the expression of c-myc protooncogene mRNA and an int-erferon inducible gene, pIFN-IND-2, mRNA(56K mRNA) has been determined.Results show that. 6-TG has strong inhibition effect and IFNs hardly have effect on HL-60 cell clonal growth. But both of them produce similar induction of terminal differentiation of HL-60 cells in NBT assay. It is very interesting that, both of them have the similar regulation effect on related gene’s mRNA expression- c-myc mRNA increased in early time, and then it decreased rapidly; 56K mRNA increased by both of them with different degrees.Results suggest that: c-myc protooncogene expression is closely related to cell differentiation and pINF-IND-2 gene might be involved with cell differentiation as well. These might be the molecular biology bases of 6-TG and IFNs.
