快速高效阴离子交换色谱填料的合成及性能研究
Study on the Synthesis and the Performance of Fast Anion-Exchange High Performance Liquid Chromatography Packing
【摘要】 将四乙烯五胺键合到含有环氧基团的交联聚苯乙烯微球表面上,经过碘甲烷反应,合成出一种非多孔的季胺化树脂,其粒度为5—9μm,交换容最为0.17meq/g(Cl)。该树脂作为高效阴离子交换色谱填料,填充到Φ8×46、Φ8×30和Φ8×27mm的柱管中,在Tris-HCl加NaCl的梯度淋洗条件下,可以实现对于蛋白质的快速分离。本文简要介绍了这种新型填料的合成,并研究了它的色谱性能。
【Abstract】 A new non-porous anion-exchange HPLC packing for fast separation of proteins has been developed.Tetraethylene pentamme was bonded on the surface of the microspherical crosslinked polystyrene containing epoxy groups,and then quaternized with methyl iodide,giving a non-porous strong anion-exchange resin with particle diameter of 5-9μm and exchange capacity of 0.17meq/g(Cl).The absence of sample diffusion in the pores that occurs in porous packing,combining with the use of short column,led to rapid separation of proteins with high efficiency and resolution.The mass recovery was 96% and the loading capacity was about 0.3mg for ovalbumin on a θ8×30mm column.
