ELISA新底物TMB终止剂的研究
STUDY ON NEW STOPPING AGENTS OF SUBSTRATE TMB IN ELISA
【摘要】 3,3′,5,5′-四甲基联苯胺(TMB)具有高度的敏感性,又无致癌性,但常规应用H2SO4终止反应,颜色会由蓝变黄,本底不断加深,阳性和阴性的对比度差,影响推广应用。本文采用了10种药物进行比较研究,发现一种理想的新终止剂——1%十二烷基硫酸钠。它能使底物显色后保持鲜蓝色不变,本底清晰如水,而且久置不会变深,阳性和阴性反应视差十分明显,既有利于目测,又可用酶联免疫检测仪于波长630nm精确判定。通过抗-HBc 108份和抗-HDV 565价标本的考核,均获得了十分满意的结果。
【Abstract】 Substrate TMB (3,3’,5,5’-Tetramethylbenzidine) is preferred in ELISA for it’s superiority: highly sensitive and non-oncogenic. Eoutinly H2SO4 is used to stop action of TMB. Neverthless, after adding H2SO4 the color changes from blue to yellow while the ground color becomes increasingly deeper as time goes on. This greatly interferes the readings. For this reason, wetry to find a more ideal stopping agent to replace H2SO4. After screening 10 agents,it was found that with 1%sodium lauryl sulfate (using syringe and fine skintest needle, 1 drop added per well) the substrate turned bright blue and kept unchanged at least for 24 hours with a clear ground. The positive and negative results were easily distinguished by naked eye or ELISA spectrophotometor at wave length of 630 nm.This kind of new stopping agent was examined in Anti-HBc (108 samples) and Anti-HDV ELISA (565 samples) with satisftory results.
