发酵法生产5’-肌苷酸的研究 Ⅰ.高产菌株的选育
STUDIES ON THE PRODUCTION OF 5′- IMP BY FERMENTATION PROCESSES I. STRAINS ISOLATION AND IMPROVEMENT FOR HIGH YIELD OF 5′-IMP
【摘要】 报道了从自然界分离产氨短杆菌(Brevibacterium ammoniagenes)进行遗传诱变,获得突变株直接发酵法生产5′—肌苷酸的研究结果.从采集的婴幼儿大便及鸟类粪便中,分离鉴定出6株产氨短杆菌.将其中的No.18和No.20用紫外线,硫酸二乙酯,亚硝基胍以多种不同方式进行诱变处理,用青霉素浓缩缺陷型.通过对2万多个单菌落的逐个检查,筛选鉴定出7株腺嘌呤缺陷型(A-),1株鸟嘌呤缺陷型(G-),12株腺嘌呤鸟嘌呤双重缺陷型(A-G-).摇瓶发酵结果,有3株A-G-产多量5′—肌苷酸,其中No.18—AG—17产量比较稳定,最高产量达12.1mg/mL。
【Abstract】 This paper reported the results of isolation and muiational works of strains of Brevibac-terium ammonlagenes and the production of 5’-IMP by fermentation processes. 6 strains of B. ammoniogenes which were isolated from feces of infants and birds have been identified. Strains No. 18 and No.20 were selected for mutagenic works with UV, DES and NTG. Penicillin were used to concentrate auxolrophic mutants. More than 20 thousands colonies were picked up from the survivors after mutagenic treatment and checked for their purine auxotro-phic mutants, and finally 7A-, 1G- and 12A-G- were obtained. All of the mutants were tested for their 5’-IMP productivities in fermentation media in shaking flasks. There were three mutants(A-G-) accumulated lurge amounts of 5’-IMP, the maximum yield was 12. 1mg/mL.
