小黑杨细胞培养及植株再生

【摘要】 本文报道了小黑杨(Populus xiaohei T.S.Hwang ex C.Wang et Tung)细胞培养及植株再生过程,包括花粉愈伤组织的诱导、继代培养、单细胞分离及培养、愈伤组织的再形成及植株再生。试验结果表明:①悬浮细胞的质量是影响细胞培养的根本因子,它主要决定于愈伤组织的质量和振荡培养时间。小黑杨三种类型愈伤组织中只有类型Ⅱ,即淡黄色,表面干燥、疏松的颗粒状愈伤组织适于制备悬浮细胞液,振荡时间一般3—5d。②用液体—固体双层培养法进行小黑杨细胞培养是一成功的尝试。MS培养基大量元素浓度、2,4-D浓度、培养基的pH值是影响细胞分裂生长的重要因子。最佳培养基为附加2,4-D 2mg/L,KT 2mg/L和2%蔗糖的改良MS培养基(大量元素减半)。③细胞培养再形成的愈伤组织转到分化培养基后能整齐一致地转变成类型Ⅰ愈伤组织,分化率和生根率均达到100%。

【Abstract】 This paper reports suspension cell culture and the process of planetlets regeneration of P. xiaohei. It includs induction and proliferation of pollen callus, isolation and culture of single cell, callus reformation and plantlet regeneration. The results are as follows: 1. The quality of suspension cells is fundamental factor affecting cell culture, and the quality is dependent on the quality of callus and the culture time of shaking. P. xiaohei has 3 types of callus, but only type Ⅱ which is yellowish, dries surface, particle state just fit for making suspension cell. The culture time of shaking is 3—5 days generally. 2. It is a successful test that using liquid-solid double layers method in cell culture of P. xiaohei. The concentration of marco-nutrient of MS medium and 2,4-D, pH of the medium are important factors affecting culture cell div ision and growth. The optimal cell culture medium of P. xiaohei is the modified MS medium (half of the macro-nutrient) supplemented with 2,4-D 2mg/L, KT 2mg/L+2% sucrose and pH 5.6—6.0. 3. After callus reformation from cell culture turn to differentiation culture, it can change to type Ⅰ callus regularly. Differentiation and rooting ratios can reach 100%
