猕猴桃细胞培养及器官分化

【摘要】 以软枣猕猴桃(Actinidia arguta)、狗枣猕猴桃(Actinidia kolomikta)和葛枣猕猴桃(Actinidia Polygama)为试材进行单细胞培养,以MS为基本培养基,附加生长素(2,4-D、NAA)和玉米素(ZT)诱导愈伤组织。结果表明,软枣猕猴桃在MS+ZT 1mg/L+2,4-D3mg/L的激素条件下,能诱导出适合于细胞培养的愈伤组织。猕猴桃游离细胞有四种繁殖方式,游离细胞培养以MS+ZT 1mg/L+2,4-D4mg/L时愈伤组织生长最好,在MS+ZT 2mg/L+NAA 0.1mg/L的激素条件下愈伤组织分化出了分生组织结节和维管组织结节。

【Abstract】 Unicellular culture in vitro of Actinidia arguta, A, kolomikta and A.polygama was carried in MS, the basic medium, and supplemented the 2,4-D or NAA and ZT to induce the callus. The results show that the callus which is suitable for cell culture can be induced under the hormone condition of MS+ZT1 mg/L+2, 4-D2 mg/L for A. arguta. There are four propagation types of the gooseberry free cell. Thecallus growth is the best under the hormone condition of MS+ZT2 mg/L+NAA0.1 mg/L for free cytes culture, but the nodes of meristem and vasscular tissue were differentiated under the hormone condition of MS+ZT2 mg/L +NAA 0.1mg/L.
