应用胶体金免疫电镜技术研究膜性肾病免疫复合物的沉积

【摘要】 膜性肾病(MN)是以肾小球上皮下免疫复合物均一沉着为特点的常见的原发性肾小球疾病。本文应用Lowicryl K 4M低温包埋的免疫电镜方法,包括双标记技术,对十例不同期MN肾小球上免疫球蛋白(IgS)成份的形成过程进行了定位、定量检测。本文结果提示,不同期MN沉积的免疫复合物其成份、数量及沉积位点是有差异的。于早期,IgG除集中在上皮下电子致密物上,还存在于足细胞间隙、上皮细胞表面,以及上皮细胞内。

【Abstract】 Membranous glomerulonephropathy is a common type of primary GN. Subepithelial dense depositsformation is its pathological feature. Ten MN patients (MN Ⅰ 4, MN Ⅱ 3, MN Ⅲ 3) were studied byultrastructual immuno-gold techinque. Indirect method was performed on the thin sections of lowtemperature embeded renal biopsy tissue with Lowicryl K4M. Our study revealed that, in early stageMN and stage Ⅱ, IgG was located not only in dense deposits of subepithelium, but also in coated pits,surface of epithelium and in the cytoplasm of epithelium. In stage Ⅲ, IgM in company with IgG waslocated in the dense deposits of G. B. M. The location and the quantity of IgS differ in differentstages of MN.
