骆驼蓬总碱体内、外对人体肝癌和胃癌瘤株的抑制作用

【摘要】 用MTT法检测骆驼蓬总碱对人肝癌BEL—7402,胃癌MGC—803和鼻咽癌CNE—2细胞株均有杀伤作用,IC50分别为4.0,17.3及24.3μg/ml,尤其肝癌细胞株最敏感。在可耐受剂量下,骆驼蓬总碱(60mg/kg)对裸鼠移植人肝癌BEL—7402和胃癌MGC—803的生长有明显的抑制作用,抑制率分别为60%和48.4%;与DDP合并用药对裸鼠异体移植肝癌BEL—7402有增效作用。

【Abstract】 Established human cancer cell lines derived from liver cancer (BEL-7402), gastric cancer (MGC-803) and nasopharyngeal cancer (CNE-2) were used in this assay. As determined by MTT assay method, the total alkaloid of peganum harmala L, was high cyto-toxic to these 3 cell lines. The IC 50’s (48 hrs incubation) were 4.0. 17.3 and 24.3 μg/ml, respectively. Among them the liver cancer cell line was the most sensitive one. Human liver cancer (BEL-7402) and gastric cancer (MGC-803) have been xenograpted subcutaneously into nure mice. At a tolerable dose level, the total alkaloid of Peganum exhibited significant antitumor activity, the inhibition rates were 60% and 48.4%, respectively for liver and gastric cancer. Synergistic antitumor effect was observed when Peganum alkaloid was combined with cisplatin (DDP).
