兔病毒性出血症病毒敏感细胞的筛选
SCREENING OF RHDV SENSITIVE CELLS
【摘要】 用兔病毒性出血症病毒(RHDV)材料,感染来源于动物及人体组织的传代细胞株和二倍体细胞共12株,经连续传代,发现其中的VeroE6、LLCMK2及L929三株细胞的分离物,经免疫荧光(IFA)及酶联免疫吸附试验(ELISA)检查,结果呈阳性反应。随带毒细胞传代次数增加.荧光亮度增强。其荧光主要在细胞核内,少数在细胞的胞浆中。用正常兔血清以及抗腺病毒3.7型、乙脑和流行性出血热兔血清染色,均为阴性。接种病料于上述三种细胞株中,可使细胞产生病变(CPE),其细胞病变分离物的滴度为10~4,这与免疫荧光结果相符合。细胞病变分离物,通过空斑试验,发现在VeroE6和L929细胞出现空斑。VeroE6细胞的分离物超薄切片,负染,电镜观察,发现细胞核内有不规则形态的包涵体。
【Abstract】 Twelve strains of passage cells and diploid somatic cells from animal and man were infected with rabbit haemorrhagic disease virus(RHDV) and continuously cultured. The separates of three strains,i. e. Vero E6, LLCMK2 and L929, showed positive reaction as determined by IFA and EL1SA. The brilliance of fluorescence was enhanced with times of passage culture, and fluorescence appeared mainly in the nucleus, less in cytoplasm. Some irregular intranuclear inclusion bodies in separates of Vero E6 strain were observed under electron microscope.
