C2-C7低级脂肪酸的气相色谱分析及其在大环内酯类抗生素结构鉴别上的应用

【摘要】 改进、建立了C2-C7低级脂肪酸及其异构体的气相色谱分析方法,对大环内酯类抗生素环状结构上的酰基进行了分析鉴别。确认抗生素2488复合物的四个单组分化合物SA1、SA2、SB1、 SB2分别为柱晶白霉素A3、A4、 A1、 A5。这一方法不但分离效果好,而且简便、准确,普遍适用于大环内酯类化合物环上酰基的结构鉴别分析。

【Abstract】 This paper dealt with an improve gas chromatographic analysis of lower fatty acids and its application on structural identification for antibiotics 2488 SA1, SA2, SB1 and SB2, with a known structure of macrolide antibiotics.Various kinds of acetyl groups and their molecular proportions of antibiotic 2488 SA1, SA2, SB1 and SB2 were identified and compared by GC analysis of their hydrolysis products, i.e. fatty acids.The mixture of C2-C7 fatty acids and their isomers could be completely resolved by using 25% PEG-20M on Shimalite(AW)(201), 60-80 mesh as stationary phase and column was operated isothermally at 170℃ and nitrogen was used as carrier gas at a flow rate of 50ml/min.Experimental results showed that antibiotic 2488 SA1, SA2, SB1 and SB2 were identical to leucomycin A3, A4, AI and A1 respectively.
